چکیده

Background: Fibrin(ogen) maintains inflammation in various disorders but has never been linked to cartilage damage rheumatoid arthritis (RA) or other forms of inflammatory arthritis. Objectives: To investigate the role fibrin deposition on integrity Methods: Fibrin knee was analyzed by immunohistochemistry RA patients and murine adjuvant-induced (AIA). In chondrocytes, fibrinogen expression ( Fgα, Fgβ, Fgγ ) procoagulant activity were evaluated qRT-PCR turbidimetry respectively. Fibrin-induced catabolic genes assessed chondrocytes. Fibrin-mediated chondro-synovial adhesion (CSA) with subsequent tears studied co-cultures human autologous synoviocytes, AIA model, MRI. The link between calcification examined stained for calcific deposits vitro fibrinogen-stimulated Results: correlated severity explants wildtype (WT) mice, while deficient (Fg -/- mice protected. Accordingly, upregulated enzymes Adamts5 Mmp13 Secondly, CSA present fibrin-rich damaged WT not Fg mice. line, cultured explants, adhered exclusively fibrin-positive degraded areas. Gadolinium-enhanced MRI joints showed contrast-enhancement along surface controls. Finally, co-localized triggered chondrocyte mineralization inducing pro-calcification Anx5, Pit1, Pc1 cytokine (IL-6). Although at a much lesser extent, we observed similar fibrin-mediated mechanisms osteoarthritis (OA). Conclusion: directly impacts via induction catabolism, mechanical stress, calcification. Potentially, is key factor occurring as secondary consequence inflammation. Disclosure Interests: None declared

Bacterial lipopolysaccharides form procollagen-endotoxin complexes that trigger cartilage inflammation and degeneration: implications for the development of rheumatoid arthritis

INTRODUCTION We have previously reported that bacterial toxins, especially endotoxins such as lipopolysaccharides (LPS), might be important causative agents in the pathogenesis of rheumatoid arthritis (RA) in an in vitro model that simulates the potential effects of residing in damp buildings. Since numerous inflammatory processes are linked with the nuclear factor-κB (NF-κB), we investigated i...

Cartilage destruction in rheumatoid arthritis.

Synovial fibroblasts represent an important source of matrix degrading enzymes mediating joint destruction in rheumatoid arthritis. In our search for new pathways for the activation of synovial fibroblasts we have detected endogenous retroviral sequences, like the line 1 element (L1). To study the functional role of L1 we transduced synovial fibroblasts with L1 constructs and showed that L1 ind...

Fibrin degradation products in rheumatoid arthritis.

The presence of fibrin in the joints, raised levels of blood fibrinogen, and an increased total daily fibrinogen turnover are well-known features of rheumatoid arthritis (Takeda, 1967) and inflammatory arthritis. A possible role for fibrin in the pathogenesis of rheumatoid arthritis, through its action as an autoimmunogen, was suggested by the production of a chronic inflammatory arthritis in r...

Patients with Active Rheumatoid Arthritis Have Lower Frequency of nTregs in Peripheral Blood

Background: Patients with rheumatoid arthritis (RA) suffer from wide ranges of autoimmune reactions in joints. The mechanism of which is generally unknown and maybe associated with Treg deregulation. Objective: To compare the frequency of nTregs in peripheral blood of patients with active rheumatoid disease with healthy individuals. Methods: Twenty five newly diagnosed patients with active RA d...
